SEO: does being listed in the Yahoo directory or DMOZ make a real difference?

It's a backlink, so of course it helps.

Well, dmoz is mirrored, so of course you will be getting more than just one backlink.

Yahoo! it really depends. You will see some traffic through the directory. If it's for a niche that converts, that traffic may pay for the listing cost. For a content site, it's probably not worth paying $200/yr, but for an E-Commerce site, it may be worth it.

Unless you have a valid google priority ID, SEO is a waste of time.

Have you found that positioning in the serps goes up along with a listing, all other things being equal?

SEO is not hard if you know what to do.

I know a adult site where the guy went from 0 to pr 6 in about 6-7 months. He had no knowledge of what seo was when he started. We just helped him and he took over and now gets thousands of free hits a day.

Not hard and everyone should do SEO by themselves or with a company, there is no better long term investment in your website.

DMOZ+Yahoo links (both of them) typically give your site a PR2-3 without any other links so I'd say its worth the effort.

Done it. When I put a new domain live, I always get the Yahoo+Dmoz combination. You need to target a category which is pretty high up in the heirchy to get this kind of effect, but its do-able for most sites.
