Can any software be setup in a multitier server achitecture? - app server?

If you are running Magento or OScommerce you have a webserver and an sql server. Where does the application server fit in? What would the point be?

Usually application servers and wan acceleration, stuff of this nature is used more for a type of environment where a company has more then 1 local office and they want to stream applications to users.
