What is the best method to getting decent looking youtube clips after they take a hatchet to the video?

I have: Premiere Pro 2, Quicktime Pro & ProCoder3 :helpme

High bitrate mp4 is always best. Just gotta maintain the 100MB limit on filesize

oh yeah don't forget to de-interlace if using a DV source ;)
