Best spam protection service?

Just wondering what everybody's opinion is on the best way to prevent spam. I'd like to sign up for one of the services that verifies the senders emails first by sending them a confirmation. But I need to be able to enter other email addresses for it to allow through no matter what, like tgp confirmation emails, sponsor newsletters, etc.

Any reccomendations on the best and most convenient service to use?
