Yea, that's the worst! You come up with a good concept and everything then you go to check on the domain and it's been taken. Then you try and tweak the name but it's never as good as the original. Then you go and check out the site to see what they are doing with it and.....nothing. So lame

You (or an agent on your behalf) could make them an offer...
With that said, making an offer is tricky - and not without risk; researching the seller, the domain, and knowledge of the domain name procedures / marketplace, often greatly helps in negotiating a reasonable purchase price.
In the meantime, especially if you want to get the project started asap, post (and/or ask around) a "Wanted" ad for the type of domain you're seeking - chances are someone out there has a suitable domain alternative for sale right now.

Yeah I hate that shit. When I am searching for domains for 90% of my new projects the domains are registered and parked. Most of the time they don't hold much value either, someone assumes that they will make a few cents per click or eventually someone will want to buy it. It is irritating for sure.
