If Romney loses I can see him running again, but it would depend on how things go over the next four years. If things go well and Obama is popular when he leaves office and a big name democrat like Hilary runs, he might not. If things don't go well he may run on the "See, I told you that you should have picked me" platform.

If Romney wins we will likely see gridlock. The democrats are likely going to hold the senate and the republicans are likely going to hold house and I was reading not too long ago about how most of the moderates from both parties are gone so it will be a congress that is very divided. The democrats will blame the republicans for sabotaging Obama and will want to return the favor and I won't be shocked to see Hilary Vs Romney in four years.
