I live in the Rhein Valley, which is about the nicest part to live here in Germany. It doesn't rain all that much here, summer and winter are mild... spring and fall are beautiful. (The Rhein Valley is where you will find a shitload of grape vineyards.)
Heidelberg is nice. It has a bit of a traditional touch that Americans like to see when they visit (my wife is American and likes Heidelberg a lot) and also has a castle for sightseeing. Heidelberg is also decent for partying because it's a college town.

Make sure you get a rental car that can go fast, because I'm sure you'll want to speed down the Autobahn while you're here. The part of the Autobahn you'd be taking from Heidelberg to Munich has some nice spots where you can really test the limits of whatever car you're driving speedwise, so I would choose to drive that part over flying it. Berlin to Frankfurt might be a bit much to drive though.
