Mashhad is the city where back in the 9th century A.D., Imam Reza was poisoned and martyred. It is a huge whole shrine for Muslims.
The Holy Shrine of Hazrat-é Ma'sumeh is the burial place of Fatima which is located only about 70 miles from Mashhad.
There are so many holy shrines in Iran. It's like invading the Vatican
Of course Mecca, located in Saudi is still the most holiest shrine for Muslims, but Mashhad is right there beside it.

3, Dome of the Rock (jerusalem)
4, Hebron (israel)
5, Mashhad (iran)
6, Qom (iran)
Iraq is not on this list. Do you really believe that Muslims will lay down and roll over if we attack one of their holiest shrines
