I'm in Rochester, NY.
We're 3-3.5 hours from Toronto driving. I'm also about 5-6 hours from Manhattan. (All depending on how fast you drive)
I'd say you'd be safe to assume that your trip will run you about 10 hours from Toronto to NYC.
