Yes it will, why not? I don't know which exactly RPG you are talking about, but the old good RPG-7 flies with a speed of 300 m/s and detonates from a rather slight barrier (a glass window, for example). If it doesn't reach the barrier, it will detonate itself after 4-6 seconds. So if you fire it off the penal, it will explode in any case even if it did not hit the target.
