If you use something like
JW Media Player (one of the many free FLV Flash players available) on your site, it has a fullscreen function, which, as long as the end user has upgraded to Flash 9 (most of the time the Flash plugin itself will ask the user to update if they haven't got the latest version), allows for fullscreen playback.
